Morphological , biochemical and physiological studies on the preservation of buffalo ( Bubalus bubalis ) spermatozoa in different modified dilutors : Effects of chloroquine - diphosphate

چکیده
A morphological, biochemical and physiological study has been made of the effects of chloroquine-diphosphate (a membrane stabilizer) on the preservation of buffalo spermatozoa in five modified dilutors designated here as Al’ A2’ A3, A4 and CAW. In almost all the five dilutors, there was significant (p< 0.01) progressive increase in the percentage of sperm abnormalities and the release of enzymes (hyaluronidase, glutamateoxaloacetic transaminase, alcohol, malate, lactate and sorbitol dehydrogenases) from spermatozoa into the media, and progressive decrease in the percentage of sperm motility and of the live sperms, with the increasing time of storage. Supplementation of these dilutors with 105 M chloroquine-diphosphate significantly (p ! 0.01) lowered the percentage of sperm abnormalities and the release of various enzymes, and increased the percentages of sperm motility and of live sperm in most of the cases. Drug capacity varied in different dilutors. It was maximum in A, (egg yolk citrate, EYC) and minimum in citric acid whey (CAW).

Effects of in vitro selenium addition to the semen extender on the spermatozoa characteristics before and after freezing in water buffaloes (Bubalus bubalis)
The aim of the present study was to investigate the effect of in vitro supplementation of selenium on fresh and frozen spermatozoa quality of buffalo (Bubalus bubalis) bulls. Five healthy buffalo bulls (5 ejaculates from each bull) were used. Each ejaculate was diluted at 37 ˚C with tris-based extender containing 0 (control), 0.5, 1, 2, 4 and 8 μg mL-1 sodium selenite and the sperm motility and...
متن کاملFactors affecting the quality of cryopreserved buffalo (Bubalus bubalis) bull spermatozoa.
Storage of buffalo (Bubalus bubalis) bull semen in the cryopreserved state is discussed in this article. Fertility rate in buffalo following artificial insemination with frozen-thawed semen is reviewed. To better understand the freezability of bubaline spermatozoa, the available data on biochemical components and the activity of specific enzymes of semen/spermatozoa are given. Moreover, the maj...
متن کاملP-142: Effects of A Synthetic Antioxidant (4-Hydroxy Tempo) Additive to The Semen Extender on the Ejaculated Spermatozoa Characteristics before and after Freezing in Water Buffaloes (Bubalus Bubalis)
Background: Semen cryopreservation is the most important section of artificial insemination programs; it allows preservation of semen fertility for a long time. Materials and Methods: The aim of the present study was to detect the effect of in vitro supplementation of 4-hydroxy Tempo on fresh and frozen spermatozoa quality of buffalo bulls. Five healthy buffalo bulls (5 ejaculates from each bul...
متن کاملP-143: Effects of In Vitro Copper Supplementation on The Ejaculated Semen Parameters in Water Buffaloes (Bubalus Bubalis)
Background: A number of studies have demonstrated membrane lipid peroxidation (LPO) as one of the causes of defective sperm function in liquid semen preserved at 4◦C and cryopreserved semen. Some attempt has been made to preserve sperm parameters, particularly sperm motility, by adding some elements and Materials to the semen before freezing. Materials and Methods: To investigate effects of cop...
متن کاملReproductive characteristics and thyroidal function in relation with season in Khuzestan buffalo (Bubalus bubalis) bulls
High ambient temperature is the major constraint on Buffalo productivity. The aim of this study was to evaluate the reproductive performance and thyroid gland function in winter and summer seasons in Khuzestan buffalo bulls. Six male indigenous buffaloes of Khuzestan with nearly the same age (2-3 years old) and weight were used. Semen and blood samples through jugular vein were collected, every...
